S400 uses 1/2.7" CCD, while S50 uses 1/1.8". So in theory, S50's picture quality is better than S400's. Smaller CCD results in more noise in picture.

S50 is a 5MP camera with full manual control, and S400 is a 4MP one without the control of shutter speed and aperture.

IMO, the only advantage of S400 is the size, which is much smaller. If size dosen't matter, S50 is definitely a winner.
