1.floor slab
2.floor slab

3.thinset layer
4.tiling
5.thinset and tiles

1.铺mud bed with mesh and Radiant Heat (optional),找平是关键。
2.thinset and tiles

Yes, the mud bed with heating elements would be the best solution.

2x4 can translate to 1.5 inch instead of 3.5 inch
sure, if you really want this way. But if I already doing the the sub floor, I will try to avoid laying it flat, I don't have the luck with the concrete floor. :D

A professional has mentioned the same thing that "如果你直接在绝缘板上铺OSB,这样绝对保证不了铺瓷砖需要的刚度". So he mentioned that there must be two layers with different types of plywood and lay down cross. I guess crossing means He also pointed out that the top layer (the second layer) of plywood has to be screwed to the bottom one but not on the sleepers.

Any reason why we shouldn't lay them flat? I even saw people used 1x4 flat as sleepers for hardwood or carpet but not for tiling floor.

Because the concrete floor won't be flat.
1.5" won't give the support you need for tiles, and you may not even get a flat subfloor. With it side way, you will have 3.5" instead of 1.5", it is much stronger.
If you can get (almost) flat support underneath, than maybe it is OK.

When the subfloor is flexible, the hardwood or carpet won't break, but the tile will.

我还是认为Platon+OSB做subfloor简单,solid, 不lost celing高度,不增加too much cost.

I'm not sure if your way of making subfloor is suitable for tiles specially in the bathroom. Laminate floor is floating floor so the subfloor doesn't need to be bound to the slab firmly. Also the slab doesn't need to be perfect level.
