Just add your wife's name to your account to make it joint... Then you can deposit your cheque. (Tell the bank she will provide signature when she returns in a few weeks.)

Why didn't you guys open a joint account in the first place?

You can open a joint account for her and deposit the check via an ATM. The only potential issue is that, when you trying to opening a joint account, the bank wants your wife to be present.
